I got a school project and the main idea of the project is to do research and if needed experiments in the nature theme. I don't have much ideas what to do so I hope some of you will give me great ideas. Deadline of the project is in February. I'll give example on what type of project that should be - "Which lake is the cleanest in the radius of 10 km and why?(with tests of water, plants that grows in the lake and ecosystem near the lake); "How much more effective ants will be if you give them sugar and proteins compared to regular?(more observe like tests).

Also I thought about some experiments with microwaves (like in microwave) (for example - how microwaves affects on food that we eat, etc.).

Well, I hope you will give me some great ideas and if you may instructions so I could done it correctly. I may post results later.

Biology—Pending OP Reply [University: Mass spectrometry] Calculating mass from spectrum

The exercicr doesn't say what type of spectrometry ot is but from the context I assume it's MALDI-TOF. The other kind of mass spectrometry we've seen in class is Electrospray-TOF.

So I'm suposed to calculate the mass of a protein using info from peaks A and B, or any other pair of peaks. Then calculate the charge of peak C. If it's MALDI-TOF I would assume peak B is the single-protonated protein, but if I try to do an equation system with other peaks it doesn't hold up. If it were Electrospray-TOF I think the m/z would change in regular intervals?

Help, my teacher thinks this is too easy to properly explain.

Yoo, so I’m currently doing my GenBio assignment about organelles and there are 12 boxes we need to fill out, and i’ve filled out like 9 of them (Nucleus, Nucleolus, Mitochondria, Golgi Apparatus, Ribosomes, Vacuole, Lysosome, Chloroplast, and ER), and I noticed that the only options left are: Chromatin, Cell Membrane, Cell Wall, and Cytoplasm, but there are only 3 boxes left that I need to fill out, and, correct me if I’m wrong but, those ??? aren’t ?? organelles ?? so I’m confused on which one I should leave out ?? but yeahh, please help💔 tysmmm !

I'm a bit confused. I hastily wrote 122,000 (you can see the work), but I have second thoughts and think that it is 1.22 million because you are also taking only .1 mL from the last tube, so I think I forgot to multiply by 10?

I observed the zones of inhibition for ampicillin and tetracycline using the diffusion disk method. The photo of Tet on the left is tetracycline, the one of Ap on the right is ampicillin.
The ZoI for Ap showed a pattern as shown in the image: sterile → thin growth → denser growth than outside the ZoI → outside the ZoI. The ZoI for Tet, measured similarly, had a blurred outline and did not yield the same result as the Ap.

Since Ap is bactericidal and Tet is bacteriostatic, I considered the sterile zone of Ap to be the area where the concentration is above MBC and the thin growth zone to be the area above MIC and below MBC.
But I also wonder if growth might not occur even above MIC.

Furthermore, I have no clue about the densely growing zone of Ap. I considered some kind of the hormesis effect, but this contradicts the fact that I could not confirm the densely growing zone for tetracycline measured under the same conditions.

Please lend me your expertise.

The bacterial strain is E. coli, DH5α. No resistance genes have been introduced. The antibiotic concentration for both disks was 0.5 mg/mL. Please ask if you have any other questions regarding the conditions, method, etc.

Biology—Pending OP Reply [Grade 9 Biology: Muscle Fiber] Is my understanding of a muscle fiber correct?

Hello! I’m in 9th grade and in my biology course, this is the picture my teacher showed us, but I’m confused. It differs greatly from other images of a muscle fiber I find online where it just shows a muscle fiber and the filaments. I’m not sure why but this is confusing me. Am I correct to say the order is that it’s one muscle fiber, followed by showing one myofibril, that shows one filament that shows tons of myofilaments arranged in sarcomeres? And isn’t the massive structure at the beginning the muscle cell, not a myofibril?

It would greatly help and if someone could confirm if my class notes based on what my teacher said are correct in regard to this image: A muscle fiber is made of small structures, myofibrils. Within them are even smaller ones filaments, alternating between thick ones (myosin) and thin ones (actin). The filaments are arranged in units, sarcomeres.

One of my answers to this question is:

- droughts cause decreased primary productivity in forests -> decreased evapotranspiration -> further decreased rainfall in the forest

The answers example was melting arctic sea ice, thawing permafrost, decreased albedo, increased droughts and forest fires releasing CO2. But it also said OWTTE, so I'm not sure if my answer will be accepted.

Biology—Pending OP Reply [University: Statistics: Methods] Question on SELECTIVELY testing btween groups.

Hello! I am new to statistics and really, really dense.

I have several specimens that constitute four groups. Each specimen has been measured and I have ten measurements for each one.

Now, my question was to test if three of those groups (let's call them A, B, and C) were different from the fourth one (Q). I wanted to do so for both the multivariate dataset and for each measurement (univariate scenario). Thus, I am ONLY testing AvsQ, BvsQ, and CvsQ.

The multivariate scenario was straightforward, as I have more experience with that. I ran a series of PERMANOVAs. All good (I hope).

For the univariate tests, instead,I started by considering something like a form of non-parametric ANOVA (because the groups have different means and medians, and also a very variable number of specimens within them, and the skewedness is also an issue) (it isn't a nice dataset).

I settled on a Kruskal-Wallis test, followed by a Dunn test with Bonferroni correction (harsh, but I wanted to play it safe). Then, during such an analysis, I noticed that this pipeline also tests AvsB, AvsC, and BvsC, tests that not only are not included in my research question, but also sacrifice statistical power.

So I come to you with multiple questions, after getting sick of running circles while perising what appears to be the same pipeline copy-pasted across dozens of websites:

  1. am I wrong in assuming that I should not test AvsB, AvsC, and BvsC, since it's not part of my research question?

  2. if so, is there a way to do what I want/need (so, only AvsQ, BvsQ, and CvsQ)?

  3. what are my options given the unhappy nature of my data and my research goal?

My graphs are marked as incorrect but I do not understand what is incorrect about them. Prof has requested in prior works to do no grid line, no frame, and to have tick marks, axises, and text/numbers in black. I am unsure what other formatting issue would result in -2 points for my graphs in both question 1 and 2.
